Tofacitinib is a small molecule inhibitor of Janus kinase (JAK) activity that prevents the JAK-dependent phosphorylation of signal transducer and activator of transcription (STAT) proteins and subsequently the STAT-induced transcription of pro-inflammatory target genes.
